Professor Carolina (Japanese: カラシナ博士 Dr. Karashina) is a Pokémon Professor that appears in the anime. She is Cynthia's grandmother. She also appears in the games and in multiple manga, but is not referred by name.
In animation
Professor Carolina is the head of the Celestic Town Historical Research Center, leading research into the Sinnoh time-space legends.
She debuted in Losing Its Lustrous! and Double Team Turnover!, where Team Galactic managed to steal the Lustrous Orb from the Research Center. Carolina was showing the Lustrous Orb to Ash and his friends at the time before Team Galactic put their plan into action. She reported the incident to the authorities, but was unaware that her close friend and fellow researcher Cyrus was the head of Team Galactic and behind the plot.
In Saving the World From Ruins!, Professor Carolina arrived at the Iron Ruins on Iron Island following the disturbance caused by Team Galactic's activities in the area. She revealed that stones that make up Iron Island and the Veilstone City meteorites were the same material. Carolina enlisted the help of Riley to continue her research.
She reappeared in The Needs of the Three!, where she was at Lake Acuity, working in combination with Professor Rowan and Gary to keep the three lake guardians safe from Team Galactic. However much of the action took place at Lake Valor, where Hunter J aided Galactic Commander Saturn in capturing all three lake guardians. Team Galactic's ultimate scheme was defeated in The Battel Finale of Legend! thanks to the group and Cynthia. Afterwards, Gary managed to secure the stolen Adamant and Lustrous Orbs, and he planned to take them back to Professor Carolina and the Celestic Town Historical Research Center.
Character
Professor Carolina is a very outspoken lady and even directly asked Ash to battle her granddaughter, Cynthia. She is very proud of her granddaughter's status as Sinnoh's Champion, telling Ash that he would have to train a lot harder if he wanted to beat Cynthia.

In the games
In the Sinnoh-based Generation IV games, the player is given an Old Charm by Cynthia to deliver to her grandmother. Once the player arrives in Celestic Town, the grandmother tells the story of the lake guardians.
In the games, once the player has faced Dialga or Palkia, she will show a picture of Dialga or Palkia, depending on which version is played. This only registers the Pokémon as "seen" in the Pokédex and does not add an entry. In Platinum, as witnessing the events at the Spear Pillar adds both Dialga and Palkia to the Pokédex, she will instead inform the player of their ability to catch Dialga and Palkia if they own their respective orbs.

Diamond & Pearl arc
Cynthia's grandmother first appeared in Suffering Psyduck, where Diamond, Pearl, and Platinum encountered her at Café Cabin, drinking her favorite drink, Moomoo Milk. In Problematic Probopass and Mad Magnezone I, she led the three through the foggy part of Route 210 to Celestic Town, telling them the legend of Sinnoh's three lakes and the lake guardians on the way.
Platinum arc
It was revealed that she was the one who had created the meteor-filled craters in Veilstone City while training to use Draco Meteor during her younger years.

Garchomp is one of Cynthia's Pokémon and her main battler. In Suffering Psyduck, Cynthia lent Garchomp to her Grandmother so that she, Diamond, Pearl, and Platinum could make it to Celestic Town safely.
Pokémon Diamond and Pearl Adventure!
Cynthia's grandmother appeared in the Pokémon Diamond and Pearl Adventure! manga in a role similar to her in the games. She is first shown in Dialga's Secret Keys, where she tried to expel members of Team Galactic from Celestic Town.
In this canon, she is the elder of the town.
